目的 研究甘氨双唑钠 (CMNa)的单剂量和多剂量药代动力学 ,以评价CMNa的蓄积性。方法 5名肿瘤病人 ,单次或每隔 1d连续 9次静脉滴注 70 0mg·m- 2 的CMNa液 ,分别在单次和第 9次静滴后设定时间点采血 ,用高效液相色谱 二极管阵列紫外法测定受试病人全血CMNa及其代谢产物甲硝唑浓度 ,对CMNa的血药浓度 时间数据进行了拟合 ,并对单剂量和多剂量的血药浓度 时间数据和药代动力学参数以及尿排泄率进行比较。结果 CMNa在病人体内分布及消除都很快 ,单剂量和多剂量静滴CMNa后血药浓度值和各药代动力学参数非常相近 ,没有明显差别。结论 甘氨双唑钠不会在肿瘤病人体内蓄积。
Objective To investigate the single dose and multiple dose pharmacokinetics of glycididazole sodium (CMNa) to evaluate the accumulation of CMNa. Methods A total of 5 tumor patients were treated with intravenous infusion of 70 0 mg · m-2 of CMNa solution either singly or every other day for 1 d. Blood samples were obtained at the set time points after single and 9 th intravenous infusion, respectively. The concentration of CMNa and its metabolite metronidazole in whole blood of patients under test were determined by diode array ultraviolet spectrophotometry. The time series data of plasma concentration of CMNa were fitted. The single-dose and multi-dose plasma concentration time data and pharmacokinetics School parameters and urinary excretion rates were compared. Results The distribution and elimination of CMNa in patients with fast, single dose and multiple doses of intravenous CMNa plasma concentration and pharmacokinetic parameters are very similar, no significant difference. Conclusion Sodium glycididazole does not accumulate in tumor patients.
