蛋白质组学是大规模、高通量、系统化从整体水平上研究蛋白质组成、活动规律及蛋白质与蛋白质相互作用的新兴学科。随着高通量核酸适配体筛选技术的发展,具有与抗体结合解离常数相似的上千种酸性、碱性、中性蛋白质的核酸适配体得以筛选。这些核酸适配体广泛应用到固定化酶解反应器、蛋白质免疫印迹、免疫共沉淀、蛋白质与蛋白质相互作用及血清(血浆)、组织、细胞等蛋白质组学研究中。本文简要综述单个核酸适配体及核酸适配体微阵列在蛋白质组学研究中的应用。
Proteomics is a large-scale, high-throughput and systematic study of the protein composition, activity rules and protein-protein interactions at an overall level. With the development of high-throughput aptamer screening techniques, thousands of acidic, basic, neutral protein aptamers with similar dissociation constants to antibodies are screened. These aptamers are widely used in immobilized enzymolysis reactors, Western blotting, co-immunoprecipitation, protein-protein interactions, and proteomics studies in serum (plasma), tissue, and cells. This article briefly reviews the application of single aptamers and nucleic aptamer microarrays in proteomics research.
