本文构建了带有HSV-TK基因的腺病毒包装质粒pACCMVTK,并获得重组腺病毒AdCMVTK,用其感染胶质母细胞瘤细胞系TJ905,研究了HSV-TK/GCV系统对该细胞系的生长影响。结果发现该系统对胶质母细胞瘤细胞系TJ905生长有明显的抑制作用,以100MOI(重复感染率)的病毒剂量感染细胞,GCV半数抑制浓度IC_(50)仅为5μmol,同时发现HSV-TK/GCV系统对TJ905细胞系有明显的旁观效应。表明该系统有治疗胶质瘤的应用前景。
In this paper, we constructed the adenovirus packaging plasmid pACCMVTK with HSV-TK gene and got the recombinant adenovirus AdCMVTK, which was used to infect the glioblastoma cell line TJ905 to study the effect of HSV-TK / GCV system on the growth of this cell line . The results showed that this system could significantly inhibit the growth of glioblastoma cell line TJ905. The cells were infected with a virus dose of 100 MOI (repetitive infection rate). The median inhibitory concentration (IC50) of GCV was only 5 μmol. HSV-TK / GCV system has obvious bystander effect on TJ905 cell line. Show that the system has the prospect of treatment of glioma.
