目的 :研究骨髓间质干细胞 (mesenchymalstemcells ,MSCs)的免疫调节作用。方法 :从胚胎股骨中分离培养间质干细胞 ,经克隆化分选出形态均一的细胞 ,用流式细胞术检测其表面标志以鉴定其纯度。将分离培养的骨髓间质干细胞接种于 96孔板 ,待其长满孔面积的 80 %~ 90 %时用直线加速器以 30GY剂量照射 ,然后与分离的外周血淋巴细胞共培养 3d ,以单独培养的淋巴细胞为对照 ,加入ConA刺激 6 8h ,MTT加入后继续温育 4h ,用酶标仪检测以观察MSCs对外周血淋巴细胞增殖反应的影响。结果 :MSCs对外周血淋巴细胞的转化有明显抑制作用 (抑制率为 2 8% )。结论 :MSCs具有抑制体外淋巴细胞增殖的作用。
Objective: To study the immunomodulatory effect of mesenchymal stem cells (MSCs). Methods: Mesenchymal stem cells were isolated and cultured from the embryonic femur. The cells were homogenized by clonal sorting and the surface markers were detected by flow cytometry to identify the purity. Bone marrow mesenchymal stem cells isolated and cultured were inoculated into 96-well plates and irradiated with a 30GY dose by a linear accelerator until 80% -90% of the area of the long hole was co-cultured with isolated peripheral blood lymphocytes for 3 days, Of the lymphocytes as a control, with ConA stimulation 6 8h, MTT added after the incubation 4h, with a microplate reader to observe the MSCs on peripheral blood lymphocyte proliferation response. Results: MSCs significantly inhibited the transformation of peripheral blood lymphocytes (the inhibition rate was 28%). Conclusion: MSCs can inhibit the proliferation of lymphocytes in vitro.
