目的研究微波消融对小鼠黑色素瘤内树突状细胞抗原提呈功能的影响。方法建立C57BL/6J小鼠B16-BL6黑色素瘤皮下移植模型,随机分成模型组、微波消融组,免疫组化检测肿瘤内CD80和CD86的表达,免疫荧光检测CD11c、MHCⅡ的表达,ELISA检测IL-12、IL-10的表达。结果微波消融组MHCⅡ、CD80和CD86表达较模型组显著增高(P<0.05);微波消融组CD11c的表达高于模型组(P<0.05);IL-12在微波消融组中高表达,而IL-10水平显著低于模型组(P>0.05)。结论微波消融可增强黑色素瘤荷瘤小鼠树突状细胞的抗原提呈功能。
Objective To study the effect of microwave ablation on the antigen presenting function of dendritic cells in mouse melanoma. Methods B16-BL6 melanoma subcutaneous transplantation model was established in C57BL / 6J mice. The models were randomly divided into model group and microwave ablation group. Immunohistochemistry was used to detect the expression of CD80 and CD86 in tumor. CD11c and MHCⅡ were detected by immunofluorescence. IL- 12, IL-10 expression. Results The expression of MHCⅡ, CD80 and CD86 in microwave ablation group was significantly higher than that in model group (P <0.05). The expression of CD11c in microwave ablation group was higher than that in model group (P <0.05) 10 levels were significantly lower than the model group (P> 0.05). Conclusion Microwave ablation can enhance the antigen presenting function of dendritic cells in melanoma bearing mice.
