目的 在中国江苏肺癌患者和对照人群中 ,检测 CYP1A1Ile/ Val、GSTM1+/ 0遗传多态性的各基因型及其联合基因型的分布 ,探讨这些基因型与肺癌个体易感性的关系。方法 配对的病例 -对照组各 10 6例 ,静脉血制备 DNA。分别应用等位基因特异性 (allele- specific,AS) - PCR和多重差别 (multi-differential,MD) - PCR,检测 CYP1A1Ile/ Val和 GSTM1+/ 0的等位基因型。结果 CYP1A1少见等位基因 Val/ Val纯合子及其与 GSTM1基因缺失纯合子 (0 / 0 )的联合基因个体 ,与对照常见基因型个体相比 ,增加了对肺癌的易感性 ,其相对危险度 (odds ratio,OR)分别为 4.0 2 (P=0 .0 3)和 9.38(P=0 .0 4) ;GSTM10 / 0纯合子及其与 CYP1A1Ile/ Val杂合子的联合基因型个体 ,显著增加了肺癌易感性 ,其 OR值分别是1.92 (P=0 .0 19)和 3.2 7(P=0 .0 1)。结论 在中国江苏人群中 ,少见基因型 CYP1A1Val/ Val,GSTM10 /0增加了个体肺癌易感性。在肺癌发生中 ,上述两种易感基因型间存在协同或相乘作用。
Objective To detect the distribution of genotypes and their associated genotypes of CYP1A1Ile/Val and GSTM1+/0 genetic polymorphisms in lung cancer patients and control population in Jiangsu, China, and to explore the relationship between these genotypes and individual susceptibility to lung cancer. Methods Paired case-control groups consisted of 106 patients each. Venous blood was used to prepare DNA. Allele-specific (AS)-PCR and multi-differential (MD)-PCR were used to detect the alleles of CYP1A1Ile/Val and GSTM1+/0, respectively. Results The single gene Val/Val homozygote of CYP1A1 rare individuals and its homozygote with GSTM1 gene homozygote (0 / 0) were associated with increased susceptibility to lung cancer and relative risk compared with common genotype individuals. (odds ratio, OR) were 4.0 2 (P=0.03) and 9.38 (P=0.04) respectively; GSTM10/0 homozygotes and their combined genotypes with CYP1A1Ile/Val heterozygote significantly increased For lung cancer susceptibility, the OR values were 1.92 (P=0.019) and 3.27 (P=0.01). Conclusion In the population of Jiangsu, China, the rare genotype CYP1A1Val/Val, GSTM10/0 increased the individual lung cancer susceptibility. In the development of lung cancer, there is synergy or multiplication between the above two susceptible genotypes.
