目的建立以石油醚洗脱杂质测定巴西绿蜂胶胶囊中总黄酮的方法。方法样品经乙醇提取,用聚酰胺粉吸附,分别用石油醚、苯不同溶剂洗去杂质;经甲醇洗脱后,在波长360 nm处,用分光光度法测定巴西绿蜂胶中总黄酮的含量。结果采用石油醚作为洗脱剂,总黄酮含量在1.9897~23.8761μg/m L范围内,与吸光度值呈良好的线性关系,浓度-吸光度回归方程为Y=0.02976X-0.00741,相关系数r2=0.99954。方法精密度为1.0%,平均回收率为96.5%。与用苯洗脱杂质结果比较,两者无显著性差异。结论该方法操作简便,重现性及稳定性好,线性范围宽,减少了苯对检测人员和环境的污染,可用于保健食品中总黄酮的测定。
OBJECTIVE To establish a method for the determination of total flavonoids in Brazil green propolis capsules by petroleum ether elution. Methods The samples were extracted with ethanol, adsorbed by polyamide powder, and washed with different solvents of petroleum ether and benzene, respectively. After the eluate was eluted with methanol, the content of total flavonoids in Brazilian green propolis was determined by spectrophotometry at the wavelength of 360 nm. Results The petroleum ether was used as eluent. The content of total flavonoids was in the range of 1.9897 ~ 23.8761μg / m L with a good linear relationship with the absorbance value. The regression equation of concentration-absorbance was Y = 0.02976X-0.00741 with correlation coefficient r2 = 0.99954 . The precision of the method was 1.0% and the average recovery was 96.5%. There is no significant difference between the two results when compared with the results of impurities eluted with benzene. Conclusion The method is simple, reproducible and stable, and has a wide linear range. It can reduce the pollution of benzene to the personnel and the environment and can be used to determine the total flavonoids in health food.
