目的研究革皮氏海参(Pearsonothuria graeffeii)皂苷对环磷酰胺所致骨髓损伤模型小鼠造血的调节作用。方法用不同剂量的革皮氏海参皂苷分别灌胃环磷酰胺所致骨髓损伤模型小鼠,采用实验血液学技术和造血祖细胞体外培养方法,检测小鼠外周血白细胞(WBC)、红细胞(RBC)、血小板(PLT)、血红蛋白(Hb)和网织红细胞(RET)数,骨髓有核细胞(BMNC),骨髓造血干细胞,粒单系祖细胞(CFU-GM),红系祖细胞(CFU-E)和巨核系祖细胞(CFU-Meg)数。结果革皮氏海参皂苷能提高小鼠外周血像中WBC、RBC、PLT、Hb和RET的数量,提高骨髓有核细胞数,促进造血干细胞的增殖和分化,增加骨髓中CFU-GM、CFU-E/CFU-E和CFU-Meg的集落生成。结论革皮氏海参皂苷对骨髓损伤模型小鼠有促进造血的作用。其作用途径可能通过调节机体的免疫功能,诱导机体产生多种造血细胞因子,促进造血干细胞的增殖,并诱导向粒单系、红系和巨核系祖细胞分化,恢复小鼠的造血功能。
Objective To investigate the regulatory effect of saponins from Pearsonothuria graeffeii on hematopoiesis in mice with bone marrow injury induced by cyclophosphamide. Methods Different doses of leather skin sea cucumber ginsenoside were intragastric administration of cyclophosphamide-induced bone marrow injury in mice, the use of experimental hematology and hematopoietic progenitor cells cultured in vitro, detection of mouse peripheral white blood cells (WBC), red blood cells (RBC PLT, RET, BMNC, CFU-GM, CFU- E) and megakaryocyte progenitor cells (CFU-Meg). Results Ginsenosides increased the numbers of WBC, RBC, PLT, Hb and RET in peripheral blood of mice, increased the number of nucleated cells in bone marrow, promoted the proliferation and differentiation of hematopoietic stem cells, increased the expression of CFU-GM and CFU- Colony formation of E / CFU-E and CFU-Meg. Conclusions Ginsenosides can promote hematopoiesis in mice with bone marrow injury. Its mechanism of action may be through regulating the body’s immune function, inducing the body to produce a variety of hematopoietic cytokines, promote the proliferation of hematopoietic stem cells, and induce differentiation to granulosa, erythroid and megakaryocyte progenitor cells and restore hematopoietic function in mice.
