目的 比较规模化与传统法制备培养基在脊髓灰质炎(脊灰)疫苗生产中对细胞产量、细胞使用率和病毒滴度的影响。方法 随机抽取5批不同方法制备的培养基,进行细胞产量、使用率、病毒滴度的平行试验。结果 规模化制备的培养基可提高细胞产量33%,提高细胞使用率29.8%,提高病毒滴度0.3个常用对数值,并使细胞污染率降低24%;结论 规模化制备培养基质量均一,批间差异小,优于传统制备法。
Objective To compare the effects of large-scale and traditional culture medium on cell yield, cell utilization and virus titer in the production of poliomyelitis (polio) vaccine. Methods Five batches of culture media were randomly selected for the parallel experiments of cell yield, usage and virus titer. Results Large-scale culture medium could increase cell yield by 33%, increase cell usage rate by 29.8%, increase virus titer by 0.3 common logarithm and decrease cell contamination rate by 24%. CONCLUSION: The difference is small, better than the traditional preparation method.
