目的研究干扰素(IFN)和白细胞介素-2(IL-2)对膀胱肿瘤细胞膜上肿瘤坏死因子受体(TNF-R)的调节作用。方法用放射配基结合分析法检测了IL-2及IFNα和γ分别作用前后膀胱癌BIU-87细胞膜TNF-R的变化。结果TNF-R的特异性结合量随着IFN与IL-2的浓度和作用时间的不同而有不同程度的增加。结论用IL-2、IFN免疫治疗后可以诱导膀胱肿瘤细胞膜TNF-R表达增加,使体液内高表达的INF可以与肿瘤细胞膜受体更好地结合以发挥生物学效应,从而杀灭肿瘤细胞。
Objective To study the regulatory effect of interferon (IFN) and interleukin-2 (IL-2) on tumor necrosis factor receptor (TNF-R) in bladder cancer cell membrane. Methods The changes of TNF-R in bladder cancer cell line BIU-87 before and after treatment with IL-2 and IFNα and γ were detected by radioligand binding assay. Results The specific binding of TNF-R increased with the concentration of IFN and IL-2 and the duration of action. Conclusions IL-2 and IFN immunotherapy can induce the expression of TNF-R in bladder tumor cell membrane to increase, so that the highly expressed INF in body fluid can better bind with tumor cell membrane receptors to exert biological effects and kill tumor cells.
