用姜片虫成虫纯化抗原(PAA,1μg/ml)预包被的PS微量反应板作快速SPA-ELISA。以本法与常规ELISA平行检测姜片虫病人滤纸干滴血40份,前者的阳性率为95.0%,后者为92.5%,二者无显著性差异(P>0.1)。以本法检测流行区献血员及非流行区健康检查者血清各40份,假阳性率分别为10%(4/40)和5%(2/40);与日本血吸虫病和长膜壳绦虫病患者血清的交叉反应率分别为12.5%(5/40)和25.0%(1/4);与肺吸虫病和肝吸虫病患者血清之间未出现交叉反应。检测81例姜片虫病患者治疗前后不同时间血清抗体的变化,提示驱虫后血清特异性抗体下降较快:在治疗后3个月,阳性反应阴转率为86.67%(65/75);半年后的阴转率高达96.00%(72/75)。研究结果表明:快速SPA-ELISA不仅敏感性高、特异性较强,而且具有考核疗效的价值。同常规ELISA相比,本法反应快速,整个试验过程仅需1小时左右。
Rapid SPA-ELISA was performed on pre-coated PS microtitre plates with zingiber adult worm antigen (PAA, 1 μg / ml). This method and conventional ELISA parallel detection of zymosis patients filter paper dry drip 40, the former positive rate was 95.0%, the latter was 92.5%, there was no significant difference between the two (P> 0.1 ). In this method, 40 serum samples were collected from blood donors and non-endemic area health check-ups in endemic areas. The false-positive rates were 10% (4/40) and 5% (2/40), respectively. The cross-reactivity rates of the patients’ serums were 12.5% (5/40) and 25.0% (1/4), respectively. No cross-reaction was found with the paragonimiasis and paragonimiasis sera. The changes of serum antibody in 81 cases of zingerosis were detected before and after treatment, suggesting that serum specific antibody decreased rapidly after de-worming: at 3 months after treatment, the positive reaction negative conversion rate was 86.67% (65/75 ); Six months after the negative rate as high as 96.00% (72/75). The results showed that rapid SPA-ELISA not only high sensitivity and specificity, but also has the value of assessment of efficacy. Compared with the conventional ELISA, the rapid response of the law, the entire test process takes only about 1 hour.
