本文报道了以东北红豆杉(TaxuscuspidataSiebetzucc。)幼嫩的茎、叶、芽为外植体的愈伤组织的诱导、生长情况以及愈伤组织中紫杉醇的含量比较。结果表明:以三个部位为外植体均能较快地诱导出愈伤组织,三个器官愈伤组织均有合成紫杉醇的能力。叶为外植体产生愈伤组织的速度较快,在第一天即有愈伤组织发生,而以芽为外植体出愈率最高,达到100%。不同部位愈伤组织的生长比较,以芽的生长率最高,其次为茎,叶的愈伤组织生长最慢,但叶愈伤组织中紫杉醇含量最高,达到0.03%,是芽或茎愈伤组织的3倍,根据紫杉醇含量和每升可产愈伤组织干重计算的每升培养基可生产紫杉醇的量,叶愈伤组织仍是芽或茎愈伤组织的2倍多。
This paper reports the induction and growth of callus from young stems, leaves and buds of Taxuscus pidata Siebetzucc as explants and paclitaxel content in callus. The results showed that all three explants could induce the callus quickly, and the three organ calluses had the ability to synthesize paclitaxel. The leaves produced faster callus for the explants and callus occurred on the first day, while the shoots with shoots as the explant had the highest rate of 100%. The growth rate of callus in different parts was higher than that of shoot, followed by stem and leaf. The callus growth was the slowest, but the content of taxol in leaf callus was the highest, reaching 0.03%, which was bud or stem. Three times the number of wounded tissues, the amount of paclitaxel per liter of culture medium was calculated based on paclitaxel content and dry weight of callus per liter, and leaf callus was still more than twice as much as shoot or stem callus.
