以 30年生东方杉的当年生顶芽、侧芽和茎段作为外植体 ,采用组织培养方法对不定芽的分化、增殖及伸长进行了研究。结果表明 :该项东方杉组织培养的最佳基本培养基为改良MS培养基。在改良MS培养基中加入了BA 10mg L可高频率诱导其不定芽的分化 ;增殖系数达 6 .5 2。芽伸长培养基为附加恶霉灵 (hy mexzal,HMI) 5 0mg L的改良MS培养基。
In this study, the adventitious shoots, lateral shoots and stem segments of 30-year-old Oriental cedar were used as explants. The differentiation, proliferation and elongation of adventitious buds were studied by tissue culture method. The results showed that the best basic medium for the tissue culture of oriental fir was modified MS medium. Addition of BA 10 mg L to modified MS medium induced the differentiation of adventitious buds with high frequency; the proliferation coefficient reached 6.52. Bud elongation medium is a modified MS medium supplemented with 50 mg L of hymexal (HMI).
