目的:观察黄荆子乙酸乙酯提出物(EVn-50)诱导人急性髓性白血病HL-60细胞分化作用。方法:体外培养HL-60细胞,MTT法测定细胞活性;瑞氏-吉姆萨染色法观察细胞分化形态学变化,间接免疫荧光显微镜观察细胞表面分化抗原CD11b表达。结果:EVn-50抑制HL-60细胞活性,其IC50值为6.29μg/mL,与全反式维甲酸(ATRA)10.35μg/mL相比较强;EVn-50具有诱导HL-60细胞向粒细胞系分化并表达分化抗原CD11b的作用,其阳性表达率呈剂量依赖性升高。结论:EVn-50具有诱导HL-60细胞分化的作用。
AIM: To observe the differentiation of human acute myeloid leukemia HL-60 cells induced by ethyl acetate propionate (EVn-50). Methods: HL-60 cells were cultured in vitro. The cell viability was measured by MTT assay. The differentiation of cells was observed by Wright-Giemsa staining and the expression of CD11b was observed by indirect immunofluorescence microscopy. Results: EVn-50 inhibited the activity of HL-60 cells with an IC50 value of 6.29 μg / mL, which was significantly higher than that of ATRA 10.35 μg / mL.
