目的 研究神经上皮肿瘤的端粒酶活性及其RNA和人端粒酶逆转录酶 (hTERT)的表达水平 ,为其临床诊断和治疗开拓思路。 方法 改良端粒重复序列扩增法及RT PCR法检测 65例神经上皮肿瘤患者肿瘤标本的端粒酶活性及其RNA和hTERT表达水平 ,并与 8例正常脑组织的标本进行对照。 结果 神经上皮肿瘤端粒酶阳性率为 61 5 % ( 4 0 / 65 ) ,hTERT阳性率为 70 8% ( 4 6/ 65 ) ,两者阳性率及其水平都随肿瘤恶性度增加而升高 (分别r=0 60 7,r=0 678,均P <0 0 1)。 结论端粒酶活性及hTERT与神经上皮肿瘤恶性程度有关 ;hTERT是调节端粒酶活性的关键酶。
Objective To study the telomerase activity and the expression of telomerase reverse transcriptase (hTERT) in neuroepithelial neoplasm and to explore ways of clinical diagnosis and treatment. Methods Telomerase activity, RNA and hTERT expression in 65 specimens of neuroepithelial neoplasm were detected by modified telomere repeat amplification (RT-PCR) and RT-PCR, and compared with that of 8 normal brain tissue. Results The positive rate of telomerase in neuroepithelial neoplasms was 61 5% (40/65) and the positive rate of hTERT was 70 8% (4 6/65). The positive rate and the level of hTERT were both increased with the increase of malignancy (R = 0 60 7, r = 0 678, respectively, P <0 0 1). Conclusion The telomerase activity and hTERT are related to the malignant degree of neuroepithelial neoplasia. HTERT is a key enzyme that regulates telomerase activity.
