目的:探讨丹参多酚酸盐治疗动脉粥样硬化的可能分子靶标。方法:将单核巨噬细胞U937设为对照组、模型组、药物处理组。其中,模型组、药物处理组以ox-LDL刺激1h,再于药物处理组中加入丹参多酚酸盐。待药物作用细胞3h后,RT-PCR法检测巨噬细胞Resistin、SRA、CD36mRNA表达水平。结果:与对照组相比,模型组Resistin、SRA mRNA表达水平均增高(P<0.01),药物处理组Resistin、SRA mRNA表达水平无差异(P>0.05);与模型组相比,药物处理组Resistin、SRA mRNA表达水平均降低(P<0.05)。各组细胞CD36 mRNA表达未见显著变化。且丹参多酚酸盐对抵抗素表达的影响作用与对清道夫受体A表达的影响作用呈正相关。结论:丹参多酚酸盐可能以抵抗素作为其药理作用的分子靶标之一,且对抵抗素表达的影响可能与清道夫受体A的表达受抑制有关。
Objective: To investigate the possible molecular targets of salvianolate treatment of atherosclerosis. METHODS: Mononuclear macrophage U937 was used as control group, model group and drug treatment group. Among them, model group and drug treatment group were stimulated with ox-LDL for 1 hour, and then Salvia polyphenolate was added to the drug treatment group. After 3 hours of treatment with the drug, the expression levels of Resistin, SRA and CD36 mRNA in macrophages were detected by RT-PCR. Results: Compared with the control group, the expression levels of Resistin and SRA mRNA in the model group increased (P<0.01). There was no difference in the expression levels of Resistin and SRA mRNA in the drug treatment group (P>0.05). Compared with the model group, the drug treatment group The expression of resistin and SRA mRNA decreased (P<0.05). There was no significant change in the expression of CD36 mRNA in each group of cells. The effect of Danshen polyphenolate on the expression of resistin and the effect on the expression of scavenger receptor A were positively correlated. Conclusion: Salvianolate may be one of the molecular targets of its pharmacological effects, and its effect on the expression of resistin may be related to the suppression of the expression of scavenger receptor A.
