目的探讨护胃散治疗急进高原胃黏膜损伤的可能作用机制。方法 350只Wistar大鼠随机分为北京组、西宁组、玛多空白组、玛多阳性对照组和护胃散高、中、低剂量组,每组50只。除北京组外,余300只大鼠空运至西宁市,玛多空白组、玛多阳性对照组和护胃散高、中、低剂量组当日继续由汽车运至玛多县。北京组、西宁组、玛多空白组灌服生理盐水2 ml,玛多阳性对照组给予荆花胃康胶丸2.4 g/(kg·d),护胃散低、中、高剂量组分别给予护胃散2.6、5.2、10.4 g/(kg·d),共计7天。各组分别于实验第1、2、3、5、7天处死10只大鼠,取胃黏膜组织进行病理观察,同时测定胃黏膜血管内皮生长因子(VEGF)和碱性成纤维生长因子(bFGF)mRNA表达。结果玛多空白组大鼠胃黏膜表面散在出血点、糜烂或浅表溃疡,局部黏膜有缺损改变;护胃散高剂量组黏膜下血管淤血、炎性渗出等较玛多空白组有较大改善。与玛多空白组同时间比较,护胃散中、高剂量组在第3、5天有不同程度的VEGF、bFGF mRNA表达升高,且护胃散高剂量组在第2、3天VEGF mRNA表达升高优于玛多阳性对照组(P<0.05或P<0.01)。结论护胃散可提高急进高原大鼠胃黏膜中VEGF及bFGF mRNA的表达,从而促进急进高原胃黏膜损伤的修复。
Objective To explore the possible mechanism of Huwei Powder in treating acute gastric mucosal injury at high altitude. Methods 350 Wistar rats were randomly divided into Beijing group, Xining group, Maduo blank group, Maduo positive control group and Huwei San high, medium and low dose group, 50 rats in each group. In addition to the Beijing group, more than 300 rats were airlifted to Maduo County by air to Xining City, Maduo blank group, Maduo positive control group and Huwei San high, medium and low dose groups. The Beijing group, the Xining group and the Maduo blank group were fed with 2 ml of normal saline. The Maduo positive control group was given Jinghuaweikang pill 2.4 g / (kg · d) Stomach powder 2.6,5.2,10.4 g / (kg · d) for a total of 7 days. The rats in each group were sacrificed on the 1st, 2nd, 3rd, 5th and 7th day respectively to observe the pathological changes of gastric mucosa. The expressions of vascular endothelial growth factor (VEGF) and basic fibroblast growth factor (bFGF) ) mRNA expression. Results The gastric mucosal surface of the Maduo blank group was scattered with hemorrhage, erosions or superficial ulcers, and the local mucosa had defects. The mucosal vascular thickening and inflammatory exudation in the high-dose Huwei San group were significantly improved compared with those in the control group . Compared with Maduo blank group, VEGF and bFGF mRNA expression increased in the medium and high dose Huaiweisan groups on the 3rd and 5th days, and the expression of VEGF mRNA increased in the high dose Hufeng San group on the 2nd and 3rd days It was superior to Maduo positive control group (P <0.05 or P <0.01). Conclusion Huweisan can improve the expression of VEGF and bFGF mRNA in gastric mucosa of rats in acute high altitude, so as to promote the repair of gastric mucosal injury in rapid plateau.
