目的对一起由多种血清型副溶血性弧菌引起的食物中毒病原菌进行毒力基因及分子分型研究。方法参照国家标准方法对事件中可疑样品进行病原菌分离鉴定,对分离的副溶血性弧菌进行血清学试验,采用荧光定量PCR检测菌株的毒力基因耐热直接溶血素基因(tdh)、耐热直接溶血素相关溶血素基因(trh)与不耐热溶血素基因(tlh),并用脉冲场凝胶电泳(PFGE)对分离到的菌株进行分子分型。结果从24份病人标本和1份剩余食品中检出副溶血性弧菌25株;血清型分布为O4:K8 4株,O3:K6 19株,O4:KUT 1株和O3:KUT 1株;分离到的25株副溶血性弧菌毒力基因检测均为tdh+tlh+trh-;PFGE聚类分析显示,不同血清型菌株分为不同的簇,同一血清型菌株的PFGE条带型间存在1~2个条带的差异,菌株间存在关联。结论这是一起由多种血清型副溶血性弧菌引起的食物中毒。
Objective To study virulence genes and molecular typing of food poisoning pathogens caused by a variety of serotypes of Vibrio parahaemolyticus. Methods According to the national standard method, the pathogenic bacteria were isolated and identified from the suspicious samples. Serum tests were carried out on the isolates of Vibrio parahaemolyticus. The heat-resistance direct hemolysin gene (tdh) Direct hemolysin related hemolysin gene (trh) and heat-labile hemolysin gene (tlh), and the isolated strains were subjected to molecular typing by pulsed-field gel electrophoresis (PFGE). Results 25 strains of Vibrio parahaemolyticus were detected from 24 patient samples and 1 remaining food. The serotypes were O4: K8, 19: O3: K6, 1: O4: KUT and 1: O3: KUT. The isolates of 25 Vibrio parahaemolyticus virulence genes were detected by tdh + tlh + trh-; PFGE cluster analysis showed that different serotype strains were divided into different clusters, PFGE bands of the same serotype strains exist Differences between 1 and 2 bands, there is a correlation between strains. Conclusion This is a food poisoning caused by various serotypes of Vibrio parahaemolyticus.
