目的探索潜伏感染状态下人巨细胞病毒(HCMV)所编码微小RNA(microRNA,miRNA),为研究其在HCMV病毒学和潜伏感染机制奠定基础。方法通过HCMV感染THP-1细胞构建HCMV潜伏感染细胞模型,采用高通量Solexa测序平台对其深度测序,并利用RNAFold等生物信息学软件进行二级结构预测和命名。结果HCMV在潜伏感染状态表达了miR-US25-2-3p、miR-US25-2-5p、miR-UL112、miR-US25-1、miR-UL22A以及长度为25 bp的编号为PC-5p-148467的miRNA,对PC-5p-148467进行系列分析,将其命名为hcmv-miR-US33as-5p。结论在潜伏感染状态下,HCMV亦可编码多种miRNA。
Objective To explore the microRNA (miRNA) encoded by human cytomegalovirus (HCMV) in latent infection and lay a foundation for studying its virological and latent infection mechanism in HCMV. Methods The HCMV latent infection cell model was constructed by infecting THP-1 cells with HCMV. The high-throughput Solexa sequencing platform was used for deep sequencing and secondary structure prediction and naming were performed by bioinformatics software such as RNAFold. Results HCMV expressed miR-US25-2-3p, miR-US25-2-5p, miR-UL112, miR-US25-1, miR-UL22A and PC-5p-148467 with a length of 25 bp in the latent infection state Of miRNAs, a series of analyzes of PC-5p-148467 were performed and named hcmv-miR-US33as-5p. Conclusions HCMV can encode a variety of miRNAs under latent infection.
