目的研究牛膝中甾酮的水煮提取工艺与含量测定方法。方法采用正交实验,筛选牛膝中甾酮的最佳水煮提取条件;用高效液相色谱法对提取物中蜕皮甾酮的含量进行测定。结果牛膝中甾酮的最佳水煮提取条件为共加入50倍量水,在80-100℃下提取2次,每次2h;以蜕皮甾酮对照品色谱峰面积y对其含量x进行回归统计,线性回归方程为:y=1.02751×106x+158195,r=0.9996,线性范围为0.406-7.308μg。结论水煮提取工艺得到牛膝提取物中蜕皮甾酮的含量为9.30%;高效液相色谱法适合于牛膝提取物中蜕皮甾酮含量的测定。
Objective To study the extraction process and determination method of the content of flavonone in Achyranthes bidentata. Methods The orthogonal extraction experiment was used to screen the best boiled extraction conditions in Achyranthes bidentata. The content of ecdysterone in the extract was determined by high performance liquid chromatography. Results The best boiled water extract condition of Achyranthes Bidentata was to add 50 times water in total, and extracted twice at 80-100°C for 2 hours each time. The peak area y of quercetin control was used to determine its content x. For regression statistics, the linear regression equation was: y=1.02751×106x+158195, r=0.9996, and the linear range was 0.406-7.308 μg. Conclusion The content of ecdysterone in extract of Achyranthes bidentata extract was 9.30%. The HPLC method was suitable for the determination of ecdysterone in Achyranthes bidentata extract.
