以宜昌百合种球诱导的鳞茎无菌系为试材,从外植体处理、固体增殖培养基优化及液体增殖培养基优化等3个方面对宜昌百合鳞茎的增殖效果进行了研究。结果表明:对外植体进行创伤处理的鳞茎诱导增殖效果极显著高于不做创伤处理;最佳固体增殖培养基为MS+6-BA2.0mg/L+NAA 0.3mg/L+蔗糖30g/L+琼脂7g/L,平均增殖倍数可达7.60倍;最佳液体悬浮增殖培养基为MS+TDZ 0.1mg/L+NAA 0.3mg/L+蔗糖30g/L,平均增殖倍数高达10.86,平均增重倍数可达6.18;在MS+6-BA 1.5mg/L+NAA 0.4mg/L+蔗糖30g/L的培养基下,液体培养的增殖效果极显著高于固体培养的诱导增殖效果。
The bulblets induced by Yichang bulbs were used as explants to study the effects of explants, the optimization of solid proliferation medium and the optimization of liquid proliferation medium. The results showed that the bulblet-induced proliferation of explants was significantly higher than that without wounding. The best solid proliferation medium was MS + 6-BA 2.0 mg / L + NAA 0.3 mg / L + sucrose 30 g / L + agar 7g / L, the average multiplication factor was up to 7.60 times. The optimal liquid suspension proliferation medium was MS + TDZ 0.1mg / L + NAA 0.3mg / L + sucrose 30g / L, the average multiplication ratio was 10.86, 6.18, respectively. Under the condition of MS + 6-BA 1.5 mg / L + NAA 0.4 mg / L + sucrose 30 g / L, the proliferation of liquid culture was significantly higher than that of solid culture.
