目的研究水提乌药与醇提乌药对急性酒精性肝损伤模型大鼠的保护作用。方法将健康雄性SD大鼠按照体重随机分为4组:正常组、模型组、水提乌药组(WYSTW)、醇提乌药组(WYCTW),每组10只。正常组,灌胃等体积蒸馏水),其余各组,均梯度灌胃给予不同浓度的白酒,每日2次,灌胃容积10m L·kg~(-1),连续10 d造模结束。自造模第l天起,给药组(均1 g·mL~(-1))分别灌胃给予相应的药物,每日1次,灌胃容积为10 m L·kg~(-1)体重,连续10 d。用全自动生化仪测定血清总胆固醇(TC)、三酰甘油(TG)、谷丙转氨酶(ALT)、谷草转氨酶(AST)、血清超氧化物歧化酶(SOD)、丙二醛(MD)水平;观察大鼠肝病理学改变,免疫组化法观察肝组织中肿瘤坏死因子-α(TNF-α)、核转录因子-κB(NF-κB)、白细胞介素-1β(IL-1β)的表达。结果与模型组比较,水提乌药组、醇提乌药组均能有效降低模型大鼠血清ALT、AST、MDA、TG含量(P<0.05);升高模型大鼠血清及肝组织SOD活性(P<0.05);能不同程度地改善模型大鼠肝组织病理学改变,抑制肝细胞中IL-1β、NF-κB、TNF-α的表达。结论水提乌药和醇提乌药组有不同程度的抗炎、抗氧化及保护肝细胞的作用。
Objective To study the protective effect of water extract and alcohol extract on acute alcoholic liver injury in rats. Methods Healthy male SD rats were randomly divided into 4 groups according to body weight: normal group, model group, WYSTW group and WYCTW group. Normal group, intragastric administration of equal volume of distilled water). The other groups were given different concentrations of white wine by intragastric gavage twice a day. The volume of intragastric administration was 10 m L · kg -1 and ended for 10 days. The levels of total cholesterol (TC), triglyceride (TG), alanine aminotransferase (ALT), aspartate aminotransferase (AST), serum superoxide dismutase (SOD) and malondialdehyde (MDA) The pathological changes of liver in rats were observed. The expression of tumor necrosis factor-α (TNF-α), nuclear factor kappa B (NF-κB) and interleukin-1β . Results Compared with the model group, both water extract group and alcohol extract group could effectively reduce the levels of ALT, AST, MDA and TG (P <0.05) in serum of model rats and SOD activity in liver tissue (P <0.05). It could improve the pathological changes of liver tissue and inhibit the expression of IL-1β, NF-κB and TNF-α in hepatocytes in different degree. Conclusions Water extract and alcohol extract have different levels of anti-inflammatory, anti-oxidation and protection of liver cells.
