目的观察黄斑明目颗粒对N-甲基-N-亚硝脲(MNU)诱导视网膜变性大鼠的保护作用。方法生后43 d的雌性SD大鼠30只随机分为3组(n=10):正常对照组、模型对照组、黄斑明目颗粒组。黄斑明目颗粒组以黄斑明目颗粒溶解后灌胃,正常对照组和模型对照组以等量蒸馏水灌胃,每日2次,连续7 d。于生后50 d,黄斑明目颗粒组和模型对照组大鼠腹腔注射MNU 40 mg.kg-1,正常对照组注射等量生理盐水。观察MNU处理后12,24,48,72 h各组大鼠视网膜电图中a波及b波的变化,并于MNU处理后72 h处死大鼠,取眼球做组织学观察。结果正常对照组大鼠不同时间点视网膜电图(ERG)a波及b波的振幅差异无显著性的意义,模型对照组在MNU处理后ERG a波及b波的振幅均呈持续性、进行性下降,黄斑明目颗粒可明显抑制ERG a波及b波振幅的降低。通过视网膜透视电子显微镜的组织学观察显示,黄斑明目颗粒组的病理组织学损害较模型对照组明显减轻。结论黄斑明目颗粒对MNU诱导的视网膜变性大鼠有保护作用,是治疗视网膜变性疾病的有效中药复方制剂。
Objective To observe the protective effect of Huangmaimingmu Granule on rats with retinal degeneration induced by N-methyl-N-nitrosourea (MNU). Methods Thirty-three female SD rats, 43 days after birth, were randomly divided into 3 groups (n = 10): normal control group, model control group and macular eyesight granule group. The macular eyesight granule group was dissolved by macular eyesight granules, then the normal control group and the model control group were given gavage with equal amount of distilled water twice daily for 7 days. At 50 days after birth, the macular eyesight granule group and the model control group were intraperitoneally injected with MNU 40 mg.kg-1, and the normal control group were injected with the same amount of normal saline. The changes of a wave and b wave in the electroretinogram of MNU were observed at 12, 24, 48 and 72 h after MNU treatment. The rats were sacrificed at 72 h after MNU treatment, and the eyeballs were observed for histological observation. Results There was no significant difference in the amplitudes of ERG a and b wave between normal control group rats at different time points. The amplitudes of ERG a and b wave in model control group were both persistent and progressive , Macular eyesight particles can significantly inhibit ERG a wave and b-wave amplitude reduction. Histopathological observation by means of the electron microscope of the retina revealed that the histopathological damage of the macular particles group was significantly reduced compared with the model control group. Conclusion Huangmaimingmu Granule has a protective effect on rats with retinal degeneration induced by MNU and is an effective traditional Chinese medicine compound preparation for treating retinal degenerative diseases.
