观察中药补肾剂──“天鼓”防治庆大霉素(GM)致耳聋作用并探讨作用机理。采用脑干诱发电位(ABR)、耳蜗铺片、血清尿素氮(BUN)及肾脏组织病理切片等方法,对补肾中药防治庆大霉素所致耳聋的听功能与肾功能变化进行同步观察。结果:周庆大霉素的豚鼠ABR阈值、血清BUN明显提高(P<0.001~0.05),内耳基底膜毛细胞数量明显减少(P<0.001);中药预防45天和治疗90天,ABR阈值、基底膜形态学及肾功能均较对照组显著改善。结论:庆大霉素引起肾损害的同时也损害内耳的功能,补肾剂可能是通过对肾脏的保护作用,从而改善内耳功能。
Observe the effects of gentamicin (GM)-induced deafness and explore the mechanism of action of traditional Chinese medicine kidney tonic agent. Using brain stem evoked potential (ABR), cochlear patch, serum urea nitrogen (BUN) and renal tissue pathology to observe the changes of the auditory function and renal function of the kidney-invigorating Chinese medicine to prevent and treat deafness caused by gentamicin. RESULTS: The threshold value of ABR and serum BUN in guinea pigs were significantly increased (P<0.001-0.05), and the number of hair cells in the basement membrane of the inner ear was significantly reduced (P<0.001); At 90 days, ABR threshold, basal membrane morphology and renal function were significantly improved compared with the control group. Conclusion: Gentamicin may cause damage to the kidneys and damage the function of the inner ear. The kidney supplement may improve the function of the inner ear by protecting the kidneys.
