目的:探讨清脑片对以皮下注射肾上腺素及冰浴刺激致大鼠急性血瘀模型血液黏度及血液流变学的影响。方法:将大鼠按体质量大小随机均匀分为空白组、急性血瘀模型组,脑络通组,大、中、小剂量清脑片组。除空白组和模型组灌胃同体积的生理盐水之外,剩下的各组灌胃相对应的药物,连续7 d,在第6天灌胃完药物60min后,除空白组大鼠,其余各组大鼠采用皮下注射肾上腺素而后进行冰水浸泡来制作大鼠急性血瘀模型。最后一次灌服药物60 min后,大鼠采用腹主动脉取血,按要求测定血液黏度及凝血酶原时间(APTT)、凝血酶时间(TT)、凝血酶原时间(PT)、血浆纤维蛋白原(FIB)凝血4项及血液流变学等指标的变化。结果:注射肾上腺素及冰浴刺激致大鼠急性血瘀模型复制成功。与模型组相比,大、中、小剂量清脑片组可显著降低高切、中切、低切全血黏度和血浆黏度(P<0.01,P<0.05),显著升高TT(P<0.01,P<0.05),显著缩短APTT(P<0.01),显著降低FIB含量(P<0.01)。结论:清脑片可以显著改善急性血瘀证模型大鼠血液黏度及血液流变学的异常变化,对血栓形成以及动脉硬化的形成有一定的预防作用。
Objective: To investigate the effect of Qingnao Tablet on the blood viscosity and hemorheology in acute blood stasis model induced by subcutaneous injection of epinephrine and ice bath. Methods: According to body mass, rats were randomly divided into blank group, acute blood stasis model group, Naoluotong group, large, medium and small dose Qingnao Tablet group. Except the blank group and the model group, the same volume of normal saline was orally administered, and the rest of the groups were orally administered with the corresponding drugs for 7 consecutive days. After the animals were orally administered for 60 minutes on the 6th day, except for the blank group, Rats in each group were injected subcutaneously with epinephrine and then soaked in ice water to make acute blood stasis model in rats. After the last administration of the drug for 60 min, blood samples were taken from the abdominal aorta, blood viscosity and prothrombin time (APTT), thrombin time (PT), plasma fibrin The original (FIB) coagulation 4 and hemorheology and other indicators of change. Results: The model of acute blood stasis was induced successfully by injection of epinephrine and ice bath stimulation. Compared with model group, large, medium and small dose Qingnaotian Tablet group could significantly reduce the high, medium, low cut whole blood viscosity and plasma viscosity (P <0.01, P <0.05), significantly increased TT (P < 0.01, P <0.05), significantly shortened the APTT (P <0.01), and significantly reduced the FIB content (P <0.01). Conclusion: Qingnao Tablet can significantly improve the abnormal blood viscosity and hemorheology in rats with acute blood stasis syndrome, and has some preventive effects on the formation of thrombosis and atherosclerosis.
