目的:探讨中药虎杖对急性肺损伤大鼠肺组织AQP-1和AQP-5 mRNA表达及免疫组化积分的影响。方法:将60只雄性Wistar大鼠随机分为正常对照组,模型组,虎杖低、中、高剂量组、强的松对照组,每组10只。造模并以虎杖煎剂治疗后,使用荧光定量RT-PCR、免疫组化方法分别测定大鼠肺组织AQP-1和AQP-5mRNA的表达以及积分情况。结果:AQP-1和AQP-5 mRNA表达显示虎杖中、高剂量组与模型组相比均明显增高,差异均有显著性(P<0.05)。虎杖低剂量组与模型组相比无显著性差异(P>0.05)。免疫组化积分结果AQP-1虎杖中剂量组最高,AQP-5高剂量组最高,低剂量组最低。3组间两两比较,差异均有统计学意义(P<0.01)。结论:虎杖可能是通过促进肺组织AQP-1和AQP-5的表达,从而起到对急性肺损伤大鼠的治疗作用。
AIM: To investigate the effect of Polygonum cuspidatum on the expression of AQP-1 and AQP-5 mRNA in lung tissue and the immunohistochemical score of lung in rats with acute lung injury. Methods: Sixty male Wistar rats were randomly divided into normal control group, model group, low, medium and high doses of Polygonum cuspidatum and prednisone control group, with 10 rats in each group. After modeling and treatment with Polygonum cuspidatum, the expression and integration of AQP-1 and AQP-5 mRNA in lung tissue of rats were detected by real-time fluorescence quantitative RT-PCR and immunohistochemistry. Results: The expression of AQP-1 and AQP-5 mRNA in Polygonum cuspidatum was significantly higher than that in model group (P <0.05). Polygonum cuspidatum low dose group compared with the model group no significant difference (P> 0.05). The results of immunohistochemistry showed that AQP-1 was the highest in the middle dose group, the highest in the AQP-5 high dose group and the lowest in the low dose group. There were significant differences between the three groups (P <0.01). Conclusion: Polygonum cuspidatum may play a role in the treatment of acute lung injury in rats by promoting the expression of AQP-1 and AQP-5 in lung tissue.
