目的:建立非水毛细管电泳法(NACE)同时测定不同批次延胡索饮片中延胡索乙素、原阿片碱、盐酸小糪碱及黄连碱含量的方法。方法:以40 mmol.L–1醋酸钠-60 mmol.L-1醋酸铵-无水甲醇缓冲液(pH 6.0)为电泳介质,未涂渍标准熔融石英毛细管(75μm×64.5 cm,有效长度56 cm)为分离通道,分离电压为25 kV,检测波长为270 nm,毛细管温度为20℃,压力进样为50 mbar,6 s。结果:4种生物碱的浓度与峰面积的线性关系良好(r>0.999 2);加样回收率为96.33%~98.10%。用此法测定了延胡索饮片中4种生物碱的含量,得到满意结果。结论:该方法简单、准确,重复性较好,可用于延胡索饮片内在质量的评价和控制。
Objective: To establish a nonaqueous capillary electrophoresis (NACE) method for simultaneous determination of tetrahydropalmatine, protopine, berberine hydrochloride and coptisine in different batches of Yanhusuo decoction. Methods: 40 mmol.L–1 sodium acetate-60 mmol.L-1 ammonium acetate-anhydrous methanol buffer (pH 6.0) was used as the electrophoretic medium without standard fused silica capillary tubes (75 μm×64.5 cm, effective length 56). Cm) is a separation channel with a separation voltage of 25 kV, a detection wavelength of 270 nm, a capillary temperature of 20°C, and a pressure injection of 50 mbar for 6 s. Results: The linear relationship between the concentration of the four alkaloids and the peak area was good (r>0.999 2); the recovery of the samples was 96.33%-98.10%. The content of four alkaloids in Yanhusuo decoction pieces was determined by this method, and satisfactory results were obtained. Conclusion: The method is simple, accurate, and reproducible. It can be used for the evaluation and control of the intrinsic quality of the dried twigs.
