人体组织中有四种不同的丙酮酸激酶同工酶表达,其中肿瘤特异性M2型丙酮酸激酶(pyruvate kinase type M2,M2-PK)在早期胚胎组织中表达并在肿瘤细胞的增殖过程中再次表达。肿瘤M2-PK通过调节细胞增殖和糖酵解调控肿瘤细胞代谢。肿瘤M2-PK在肿瘤细胞中主要以二聚化形式存在,M2-PK与某些肿瘤蛋白之间的相互作用使其二聚化并且在二聚体和四聚体之间相互转变,从而使肿瘤细胞在变化的氧和营养供应的环境下生长。肿瘤M2-PK作为可肿瘤标志物用于恶性肿瘤的诊断和治疗监测。
There are four different pyruvate kinase isoenzymes in human tissues, in which the tumor-specific pyruvate kinase M2 (M2-PK) is expressed in early embryonic tissues and again in the course of tumor cell proliferation expression.
