目的观察补充一定剂量的维生素A(VA)和维生素D3(VD3)对大鼠淋巴细胞增殖活性和单核细胞吞噬功能的影响。方法取初断乳Wistar大鼠40只,雌雄各半,随机分为对照组、VA组、VD3组和VA+VD3组,均给予基础饲料饲养。对照组按大鼠体质量用大豆色拉油(1mL/kg)灌胃,摄入VA和VD3均为大鼠生理需要量(VA剂量为278U/(kg.d),VD3剂量为20U/(kg.d);用大豆色拉油将VA、VD3稀释成不同浓度油剂后按1mL/kg体质量给予大鼠灌胃:VA组为1 390U/(kg.d);VD3组为200U/(kg.d);VA+VD3组为VA 1 390U/(kg.d)+VD3200U/(kg.d);实验为4周。采用CCK-8法和CD-14标记单核细胞后用流式细胞术分别检测淋巴细胞增殖活性和单核细胞荧光强度。结果 4周后VD3组大鼠淋巴细胞增殖活性明显增强,与对照组比较,差异有显著性(F=6.72,q=6.04,P<0.05)。VA组、VD3组和VA+VD3组单核细胞吞噬功能与对照组比较明显升高,差异均有显著性(F=5.26,q=3.65~4.90,P<0.05)。结论补充生理剂量5倍的VA或(和)10倍的VD3可有效地增强大鼠淋巴细胞增殖活性和单核细胞吞噬活性。
Objective To observe the effects of vitamin A (VA) and vitamin D3 (VD3) supplementation on lymphocyte proliferation activity and monocyte phagocytosis in rats. Methods Forty weaned Wistar rats were randomly divided into control group, VA group, VD3 group and VA + VD3 group, all fed with basal diet. The rats in the control group were intragastrically infused with soybean salad oil (1 mL / kg body weight), and the VA and VD3 intake were all physiological requirements (VA dose 278U / (kg.d), VD3 dose 20U / kg .d); VA and VD3 were diluted with soybean salad oil into different concentrations of oil and then given to rats at a dose of 1 mL / kg body weight: 1 390 U / (kg.d) in VA group and 200 U / (kg .d). The VA + VD3 group was VA1 390U / (kg.d) + VD3200U / (kg.d) for 4 weeks.The monocytes were labeled with CCK-8 and CD- The proliferative activity of monocytes and the fluorescence intensity of monocytes were detected respectively.Results After 4 weeks, the proliferation activity of lymphocytes in VD3 group was significantly higher than that in control group (F = 6.72, q = 6.04, P <0.05 ) .The phagocytic function of monocytes in VA group, VD3 group and VA + VD3 group was significantly higher than that in control group (F = 5.26, q = 3.65-4.90, P <0.05) 5 times the VA or (and) 10 times the VD3 can effectively enhance the rat lymphocyte proliferation activity and monocyte phagocytic activity.
