目的建立地塞米松(DEX)诱导的L6骨骼肌细胞胰岛素抵抗模型,探讨葛根芩连汤及其主要成分(葛根素、小檗碱、黄芩苷)对L6骨骼肌细胞胰岛素抵抗模型葡萄糖消耗量的影响。方法将诱导分化好的L6肌细胞按106个·ml-1接种于96孔板内,以MTT法检测葛根芩连汤含药血清对L6肌细胞增殖的影响;以1μmol/L DEX诱导L6肌细胞24 h产生胰岛素抵抗,予以不同浓度的葛根芩连汤含药血清及其主要成分(葛根素、小檗碱、黄芩苷)进行干预,100 nmol/L胰岛素刺激30min,采用葡萄糖氧化酶法检测L6肌细胞的葡萄糖消耗量。结果葛根芩连汤含药血清对L6肌细胞增殖无显著影响。与正常组比较,模型组葡萄糖消耗量明显下降;葛根芩连汤含药血清(5%、10%、15%)、葛根素(1,10,100μmol·L-1)、小檗碱(1μmol/L)和黄芩苷(1,10,100μmol·L-1)能显著增加L6骨骼肌细胞胰岛素抵抗模型葡萄糖消耗量(P<0.05)。结论葛根芩连汤及其主要成分(葛根素、小檗碱、黄芩苷)可调节L6骨骼肌细胞胰岛素抵抗模型糖代谢,进而可改善肌细胞胰岛素抵抗。
Objective To establish a model of L6 skeletal myocyte insulin resistance induced by dexamethasone (DEX) and explore the effects of Gegen Qinlian Decoction and its major components (puerarin, berberine and baicalin) on the glucose consumption of L6 skeletal muscle insulin resistance model influences. Methods The differentiated L6 myocytes were inoculated into 96-well plates at a concentration of 106 · ml-1. The effects of Gegen Qinlian decoction-containing serum on the proliferation of L6 myocytes were detected by MTT assay. L6 muscle cells were induced by 1μmol / L DEX The cells were exposed to insulin for 24 h. Different concentrations of Puerarin, Scutellariae Radix were treated with different concentrations of puerarin, berberine and baicalin, and then stimulated with 100 nmol / L insulin for 30 min. Glucose oxidase L6 muscle cell glucose consumption. Results Gegen Qin Lian Decoction serum had no significant effect on the proliferation of L6 myocytes. Compared with the normal group, the glucose consumption of the model group decreased significantly. The contents of Puerarin, Decoction of Puerarin, L) and baicalin (1, 10, 100μmol·L-1) significantly increased glucose consumption in L6 skeletal muscle insulin resistance model (P <0.05). Conclusion Gegen Qinlian Decoction and its main components (puerarin, berberine, baicalin) can regulate glucose metabolism in insulin resistance model of L6 skeletal muscle cells, and then improve insulin resistance in muscle cells.
