目的从分子水平鉴别濒危药用植物毛黄堇和石生黄堇的差异。方法分别从毛黄堇和石生黄堇的叶片中提取DNA,以核基因组通用引物为引物进行扩增,扩增产物经纯化后对PCR产物采用直接测序法进行测序。结果获得ITS及5.8 S rDNA完全序列,分别为毛黄堇561 bp、石生黄堇552 bp;二者的ITS序列差异呈显著性,其中ITS1差异性达16.28%,ITS2差异性达21.21%。结论 ITS序列可有效地鉴别毛黄堇和石生黄堇,并可广泛应用于中药材的鉴别。
Objective To identify the difference between the endangered medicinal plants, Epimedium brevicornum and Epimedium, from the molecular level. Methods DNA was extracted from the leaves of Phellinus igniarius and Pistacia chinensis respectively. The universal primer of nuclear genome was used as primer to amplify the amplified product. The PCR products were sequenced by direct sequencing. ITS and 5.8S rDNA sequences were obtained, which were 561 bp in phellodendron chinense and 552 bp in phellodendron chinense, respectively. The ITS sequences of the two sequences were significantly different, including 16.28% in ITS1 and 21.21% in ITS2. Conclusion ITS sequence can effectively identify the yellow pansy and stone phellinus, and can be widely used in the identification of Chinese herbal medicines.
