作者用双层琼脂培养技术,测定了32例血液学检查正常人的血浆集落刺激活性(CSA)。双层培养体系的上层为1ml.3%的琼脂McCoy!s 5A培养液,内含15%健康产妇胎盘血清和1×10~5个人骨髓有核细胞。底层由1.5ml 0.5%的琼脂营养层组成,内含15%健康产妇胎盘血清和0.1~0.75ml无菌待测血浆,即底层中的待测血浆浓度为6.7%~50%。对照组的底层琼脂中含有0.75ml199培养液。于37℃下培养7~10天后计数不同血浆浓度所刺激产生的集落数(CFUc)。CFUc分为小(<40个细胞),中(40~100个细胞),大(>100个细胞)三种。以血浆浓度为横座标,CFUc数为纵座标,作出剂量-效应曲线。为便于与其他类似实验作比较,可将测得结果作准标化处理,即以最高CFUc数为100%,求出不同浓度血浆所刺激形成的CFUc数为最高数的百分数。测定结果:有69%的血液学正常人可检查出CSA。底层中血浆浓度在从6.7%增加到50%时,所形成的CFUc数及其体积随之增加(血浆为0.1,0.3
The authors measured the colony-stimulating activity (CSA) in 32 hematological normal subjects using double-agar culture. The upper layer of the double-layer culture system is 1 ml of agar McCoy’s 5A liquid containing 15% of healthy maternal placental serum and 1 x 10-5 human bone marrow nucleated cells.
