目的:观察灰树花多糖对化疗相关性疲劳(CIF)模型疲劳程度的改善情况,并初步探讨其可能的机制。方法:6~8周龄SPF级C57BL/6小鼠60只随机分为4组,即空白组,模型组,灰树花多糖低剂量组(5 mg·kg~(-1))和灰树花多糖高剂量组(10 mg·kg~(-1)),每组15只。将除空白组以外的小鼠腹腔注射5-氟尿嘧啶(5-Fu,40 mg·kg~(-1)),每天给药1次,连续5 d,同时灰树花多糖低剂量组和灰树花多糖高剂量组给予不同剂量灰树花多糖灌胃,空白组和模型组给予等量生理盐水灌胃,期间测量体重和抓力变化。第15天处死动物取材测血尿素氮、血乳酸和肝糖原含量及腓肠肌p38丝裂原活化蛋白激酶(p38MAPK)mRNA和磷酸化p38 MAPK(p-p38 MAPK)的表达情况。结果:低剂量和高剂量的灰树花多糖均能明显增强CIF小鼠的抓力(P<0.01);低剂量和高剂量灰树花多糖均可明显降低CIF小鼠运动后的血尿素氮值(P<0.05);高剂量灰树花多糖可以明显降低CIF小鼠运动后的血乳酸值(P<0.01);低剂量和高剂量灰树花多糖均可明显降低腓肠肌p38 MAPK mRNA和p-p38 MAPK蛋白的表达水平。结论:灰树花多糖可以有效改善化疗相关性疲劳,可能与下调p38 MAPK基因与p-p38 MAPK的表达有关。
Objective: To observe the effect of Grifola frondosa polysaccharide on fatigue of chemotherapy-related fatigue (CIF) model and to explore its possible mechanism. Methods: Sixty SPF C57BL / 6 mice (6-8 weeks old) were randomly divided into four groups: blank group, model group, low dose Grifola frondosa (5 mg · kg -1) High-dose polysaccharides group (10 mg · kg -1), 15 rats in each group. Mice except the blank group were intraperitoneally injected with 5-fluorouracil (40 mg · kg -1) once daily for 5 days, while the Grifola frondosa low dose group and the Grifola frondosa The high-dose polysaccharides group was given different doses of Grifola frondosa gavage, the blank group and model group were given the same amount of saline gavage, weight and grip during the measurement changes. The contents of blood urea nitrogen, blood lactic acid and hepatic glycogen and p38 mitogen-activated protein kinase (p38 MAPK) mRNA and p38 MAPK expression in gastrocnemius muscle were measured on the 15th day. RESULTS: Both low dose and high dose of Grifola fritillaria polysaccharides could significantly enhance the grasping ability of CIF mice (P <0.01). Both low dose and high dose Grifola frondosa polysaccharide could significantly decrease the blood urea nitrogen (P <0.05). High dose Grifola frondosa polysaccharide could significantly decrease the blood lactic acid (P <0.01) after exercise in CIF mice. Both low dose and high dose Grifola frondosa polysaccharide could significantly decrease the expression of p38 MAPK mRNA and p -p38 MAPK protein expression level. Conclusion: Grifola frondosa polysaccharide can effectively improve the chemotherapy-related fatigue, which may be related to the downregulation of p38 MAPK gene and the expression of p-p38 MAPK.
