本研究观察TCR独特型DNA疫苗诱导BALB c小鼠抗肿瘤免疫反应的情况。用CEM淋巴瘤细胞系和BALB c小鼠作模型 ,RT PCR扩增CEM细胞系特异性重排的TCRVβ基因 ,克隆到真核表达载体pcDNA3中作为DNA疫苗。用肌内注射法免疫小鼠 ,间接免疫荧光法检测小鼠抗体生成和用MTT法测定CTL活性以检测抗独特型的细胞免疫。结果发现 ,接种DNA疫苗后第 4周开始小鼠血清中即可检测到特异性抗独特型抗体 ,抗体滴度在第 6周达高峰 ,应用IL 6为免疫佐剂可使抗体滴度明显增高。上述结果表明 ,TCR独特型DNA疫苗可以诱导小鼠产生抗淋巴瘤细胞的独特型特异性抗体 ,应用IL 6为免疫佐剂可使这种反应明显增强
In this study, the TCR idiotypic DNA vaccine was used to induce anti-tumor immune responses in BALB c mice. Using the CEM lymphoma cell line and BALB c mice as a model, RT-PCR was used to amplify the specific rearranged TCRVβ gene of CEM cell line and cloned into the eukaryotic expression vector pcDNA3 as a DNA vaccine. Intramuscular injection was used to immunize mice, indirect immunofluorescence was used to measure antibody production in mice, and MTT assay was used to measure CTL activity to detect anti-idiotypic cellular immunity. The results showed that specific anti-idiotypic antibodies could be detected in the serum of the mice starting 4 weeks after the vaccination of the DNA vaccine. The titer of antibody peaked at the 6th week, and the antibody titer was significantly increased by using IL 6 as an adjuvant. . The above results indicate that the TCR idiotypic DNA vaccine can induce mice to produce idiotype-specific antibodies against lymphoma cells. The use of IL 6 as an immunoadjuvant can significantly enhance this response.
