目的:探讨胃泌素释放肽(GRP)对高转移人肝癌细胞MHCC97H迁移和上皮间质转化(EMT)的作用。方法:利用外源性的GRP刺激MHCC97H细胞,细胞三维培养观察细胞形态的变化,Westernblot检测EMT标志蛋白E-cadherin和vimentin的表达,Transwell迁移分析检测肝癌细胞的迁移数量的变化。结果:与对照组相比较,GRP刺激的人高转移肝癌细胞MHCC97H向间叶细胞的形态转变,使上皮细胞表型标记物E-cadherin蛋白表达下降,间叶细胞表型标记物vimentin蛋白表达升高,细胞迁移数量明显增多。其作用能被GRP受体阻滞剂PD176252所减弱。结论:GRP能够诱导人高转移肝癌细胞MHCC97H发生EMT,促进肝癌细胞的迁移,该作用是通过GRP结合GRPR实现的。
Objective: To investigate the effect of gastrin-releasing peptide (GRP) on MHCC97H migration and epithelial-mesenchymal transition (EMT) in highly metastatic human hepatoma cells. Methods: MHCC97H cells were stimulated with exogenous GRP. The cell morphology was observed by three-dimensional culture. The expression of EMT markers E-cadherin and vimentin were detected by Western blotting. The migration of hepatoma cells was detected by Transwell migration assay. Results: Compared with the control group, the morphology of MHCC97H cells in GRP-stimulated human hepatocellular carcinoma cells changed to mesenchyme. The expression of E-cadherin, a marker of epithelial cell phenotype, decreased and the expression of mesenchymal marker vimentin increased High, the number of cell migration increased significantly. Its role can be GRP receptor blocker PD176252 weakened. CONCLUSION: GRP can induce EMT in human highly metastatic hepatocellular carcinoma cell line MHCC97H and promote the migration of hepatocellular carcinoma cells. This effect is mediated by GRP combined with GRPR.
