华蟾素为中华大蟾蜍皮水煮醇沉制剂,临床具显著抗肿瘤活性。本文首次采用高效液相色谱串联高分辨傅立叶变换离子回旋共振质谱(HPLC-DAD-FT-ICR-MS)方法,针对华蟾素中主要抗肿瘤活性成分蟾毒配基类化合物进行了分析,旨在阐明华蟾素抗肿瘤活性效应物质基础。该研究首先采用二氯甲烷-硅胶固相萃取法对华蟾素中蟾毒配基类成分进行富集,根据化合物的UV及FT-ICR-MS高分辨质谱的一级、二级质谱数据,结合文献,比较分析了华蟾素中的主要蟾毒配基类成分的化学组成,共推导出33个蟾毒配基类成分,其中8个成分通过与对照品比对,准确鉴定了结构。该研究基本阐明了华蟾素中主要蟾毒配基类成分,为华蟾素的进一步药效学研究奠定了物质基础。
Cinobufagin is a Chinese toad skin water boiled alcohol preparation, clinically significant anti-tumor activity. In this paper, the method of HPLC-DAD-FT-ICR-MS coupled with high performance liquid chromatography tandem mass spectrometry (HPLC-DAD-FT-ICR-MS) In elucidating the cinobufotalin antitumor activity effect substance basis. In this study, we first used the dichloromethane-silica gel SPE method to enrich the bufogenin-containing components of cinobufacini. According to the UV and FT-ICR-MS high-resolution MS data of the first and second MS spectra, Based on the literature, the chemical constituents of the major toads in the cinobufacini were comparatively analyzed. A total of 33 toadstool ligands were deduced, of which eight were identified by comparison with the control. This study basically elucidated the major components of the bufogenin in cinobufacini, which laid the material foundation for the further pharmacodynamic study of cinobufacini.
