目的:检测正常、妊娠与肌瘤组织的人子宫平滑肌细胞(HMC)胞浆和胞膜中蛋白激酶C(PKC)的比活性,比较其异同.方法:根据同位素放射免疫结合实验技术,应用同位素标记的[?-32P]ATP测定PKC活性.结果:基础状态下,HMC胞膜PKC比活性为(4.15±0.24)NMOL/GPR?MIN-1,高于胞浆(1.88±0.37)NMOL/GPR?MIN-1(P<0.05);肌瘤组织胞膜PKC比活性为(4.80±0.57)NMOL/GPR?MIN-1,高于胞浆(2.75±0.63)NMOL/GPR?MIN-1(P<0.05);妊娠组织胞膜PKC比活性为(3.98±0.48)NMOL/GPR?MIN-1,高于胞浆(3.24±0.69)NMOL/GPR?MIN-1(P<0.05);且肌瘤与妊娠时HMC的PKC总活性高于正常组织.10μMOL/L乙酰胆碱(ACH)、4U/L催产素(OT)可使HMC的胞膜PKC比活性升高、胞浆PKC比活性轻度下降、总活性增加;100NMOL/L佛波酯(TPA)则可使HMC的胞膜PKC比活性升高、胞浆PKC比活性下降、总活性基本不变.正常组织和肌瘤组织以TPA作用最强,妊娠组织以OT作用最“,”Objective: Our paper was designed to determine the protein kinase C (PKC) activity in human myometrium cell (HMC) plasma and cytomembrane of normal, pregnant, myoma and to analyze the physiological and pathological mechanism of PKC signal transduction. Methods: According to the technology of isotope radioimmune combination, the PKC activity was assayed by measuring the incorporation of 32 P from 32 P ATP into protamine. Results: For the normal controls, the PKC activity in HMC cytomembrane was (4.15±0.24) nmol/g Pr·min -1 , which was higher than that in plasma (1.88±0.37) nmol/g Pr·min -1 (P<0.05). For the myoma group, the PKC activity in HMC cytomembrane was (4.80±0.57) nmol/g Pr·min -1 , which was higher than that in plasma (2.75±0.63) nmol/g Pr·min -1 (P<0.05). For the pregnant group, the former was also higher than that latter. These two values were 3.98±0.48 nmol/g Pr·min -1 and (3.24±0.69) nmol/g Pr·min -1 , respectively (P<0.05). The total PKC activities in myoma and pregnant myometrium were all higher than that in normal myometrium. Treated with 10 μmol/L acetylcholine (ACh), 4 U/L oxytocin (OT), the PKC activities increased in all HMC cytomembrane, but decreased slightly in all HMC plasma, the total PKC activities increased. Treated with 100 nmol/L 12 O tetradecanoylphorbol 13 acetate (TPA), the total PKC activities increased in all HMC cytomembrane, decreased in all HMC plasma, but the total activities remained unchanged. Furthermore, the action of TPA was highest in normal and myoma myometrium, and the action of OT was highest in pregnant myometrium. Conclusion: Under physiological and pathological conditions, the PKC activities in HMC and its reactions to activitors were different, the PKC total activities in tumor and pregnant myometrium were higher than those in normal myometrium. The roles of PKC in cell growth and differentiation might be different.
