目的 探讨猪鼻支原体抗原对白血病细胞增殖和凋亡的影响及机制。方法 以MTT法测定猪鼻支原体抗原对白血病细胞系NB4 、HL 6 0及K5 6 2增殖的影响 ;琼脂糖凝胶电泳检测K5 6 2细胞凋亡 ;TUNEL法及二苯胺法定量检测细胞凋亡 ;同时观察猪鼻支原体抗原对K5 6 2细胞Bcl 2 Bax表达的影响。结果 猪鼻支原体抗原呈浓度依赖性抑制 3种白血病细胞的增殖 ;在有效浓度的抗原作用下 ,DNAladder显示K5 6 2细胞以凋亡方式死亡 ;TUNEL法获得凋亡率为 38.6 5 % ,显著高于正常对照组 (10 .2 3% ) ;二苯胺法获得凋亡率为 2 5 .0 1% ,显著高于正常对照组 (16 .4 9% ) ;与对照组比较 ,Bcl 2表达降低 ,Bax表达升高 ,Bcl 2 Bax比值下降 (为 0 .6 5 91)。结论 猪鼻支原体抗原呈浓度依赖性抑制白血病细胞的增殖 ;猪鼻支原体抗原通过诱导K5 6 2细胞凋亡的方式抑制其增殖 ,可能是通过降低Bcl 2、升高Bax而实现的。
Objective To investigate the effect of Mycoplasma hyopneumoniae antigen on the proliferation and apoptosis of leukemia cells and its mechanism. Methods MTT assay was used to determine the effects of Mycoplasma hyopneumoniae antigens on the proliferation of leukemic cell lines NB4, HL 60 and K562; the apoptosis of K562 cells was detected by agarose gel electrophoresis; the apoptosis was detected by TUNEL and diphenylamine; Simultaneously, the effect of Mycoplasma hyopneumoniae antigen on the expression of Bcl 2 Bax in K5 6 2 cells was observed. Results Mycoplasma hyorhinogen antigen inhibited the proliferation of three kinds of leukemia cells in a concentration-dependent manner. DNAladder showed that K562 cells died by apoptosis under the effective concentration of antigens. The apoptosis rate was 38.6 5% by TUNEL method, which was significantly higher than The normal control group (10.23%); diphenylamine method to obtain the apoptosis rate was 25.0%, significantly higher than the normal control group (16.49%); compared with the control group, Bcl 2 expression decreased, Bax expression increased, Bcl 2 Bax ratio decreased (0.56). Conclusion Mycoplasma hyorhinis antigen inhibits the proliferation of leukemia cells in a concentration-dependent manner. Mycoplasma hyopneumoniae inhibits the proliferation of K562 cells by inducing the apoptosis of K562 cells in a dose-dependent manner, possibly by decreasing Bcl 2 and increasing Bax.
