目的:研究不同炮制方式的何首乌中大黄素与大黄素甲醚的最佳提取工艺。方法:采用HPLC来测定何首乌生药、制何首乌、豆制何首乌中大黄素与大黄素甲醚的含量,以成分含量为指标,用响应面法建立甲醇浓度、料液比、提取时间3个因素的回归模型,优化提取工艺。结果:最佳提取工艺条件为纯甲醇,料液比1:30,提取时间1.5 h。在此条件下制何首乌中大黄素的含量为1.63 mg·g~(-1)生药,大黄素甲醚含量为0.95 mg·g~(-1)生药。结论:制何首乌中大黄素与大黄素甲醚含量较高。优化后的工艺适用于何首乌不同炮制品中游离大黄素和大黄素甲醚的提取测定。
Objective: To study the best extraction process of emodin and physcion in different ways of processing Polygonum multiflorum Thunb. Methods: HPLC was used to determine the contents of emodin and physcion in Polygonum multiflorum Thunb. And Polygonum multiflorum Thunb., And the content of methanol was used as the index to establish the three parameters of methanol concentration, solid-liquid ratio and extraction time by response surface methodology Regression model, optimize the extraction process. Results: The optimum extraction conditions were pure methanol, the ratio of material to liquid was 1:30 and the extraction time was 1.5 h. Under this condition, the content of emodin in Polygonum multiflorum was 1.63 mg · g -1 crude drug, and the content of physcion was 0.95 mg · g -1 crude drug. Conclusion: The content of emodin and physcion in Polygonum multiflorum is higher. The optimized process is suitable for the determination of free emodin and physcion in different processed products of Radix Polygoni Multiflori.
