目的:探讨肝细胞生长因子(HGF)及肿瘤坏死因子-α(TNF-α)在妊娠期糖尿病(GDM)Wistar大鼠胎盘中的表达及与胰岛素抵抗的关系。方法:应用高糖、高脂饲料喂养健康Wistar雌性大鼠56只和Wistar雄性大鼠20只,将雌性大鼠随机分为两组,实验组(28只)给予高糖、高脂饲料喂养5周;对照组(28只)给予普通饲料喂养5周。两组采集鼠尾静脉血测空腹胰岛素及空腹血糖水平,确定动物模型。于实验第6周给予合笼受孕,受孕后第20天采集鼠尾静脉血,检测两组空腹血糖及空腹胰岛素水平;应用RT-PCR法检测两组Wistar大鼠胎盘组织中HGF m RNA及TNF-αm RNA的相对表达情况。结果:应用高糖、高脂饲料喂养Wistar大鼠5周,测定大鼠空腹胰岛素及空腹血糖水平均异常,提示造模成功;实验组Wistar大鼠鼠尾静脉血清空腹血糖及空腹胰岛素水平明显高于对照组(P<0.05);实验组Wistar大鼠胎盘组织中HGF m RNA表达水平低于对照组(P<0.05);实验组Wistar大鼠胎盘组织中TNF-αm RNA表达水平高于对照组(P<0.05);结论:胎盘组织中HGF低表达及TNF-α高表达与GDM胰岛素抵抗密切相关。
Objective: To investigate the expression of hepatocyte growth factor (HGF) and tumor necrosis factor-α (TNF-α) in the placenta of gestational diabetes mellitus (GDM) Wistar rats and its relationship with insulin resistance. Methods: 56 healthy Wistar female rats and 20 Wistar male rats were fed with high-sugar and high-fat diet. The female rats were randomly divided into two groups. The experimental group (28) was fed with high glucose and high fat diet 5 Week; control group (28) fed normal feed for 5 weeks. Two groups of tail vein blood collected from fasting insulin and fasting blood glucose levels, to determine the animal model. The rats were sacrificed at the 6th week of the experiment. Twenty days after conception, the tail vein blood was collected and the fasting blood glucose and fasting insulin levels were measured. The levels of HGF mRNA and TNF in the placenta of Wistar rats were detected by RT-PCR. -αm RNA relative expression. Results: Wistar rats were fed with high-sucrose and high-fat diet for 5 weeks. The levels of fasting insulin and fasting blood glucose in rats were all abnormal, which suggested that the model was successful. The fasting blood glucose and fasting insulin in the tail vein of Wistar rats were significantly higher (P <0.05). The levels of HGF mRNA in placenta of Wistar rats in experimental group were lower than those in control group (P <0.05). The levels of TNF-αmRNA in placenta of Wistar rats in experimental group were higher than those in control group (P <0.05). Conclusion: The low expression of HGF and the high expression of TNF-α in placenta are closely related to the insulin resistance of GDM.
