目的 研究电磁脉冲 (EMP)对犬淋巴细胞的损伤效应 ,并探讨其作用机理。方法 用酸性磷酸酯酶细胞化学方法检测T、TH 和TS 淋巴细胞 ,用原位末端标记和免疫细胞化学技术分析淋巴细胞凋亡和Bcl 2、Bax蛋白的改变。结果 (2 - 12 )× 10 4 V/m的场强能使外周血T细胞及其亚群出现明显降低 ;凋亡淋巴细胞随照射剂量的增加而明显升高 ;不同剂量照射后 10天 ,淋巴细胞Bax蛋白的表达明显增强 ,与淋巴细胞凋亡呈现出比较好的相关性 ,而Bcl 2蛋白显著降低。结论 首次证实一定场强的EMP确实能损伤淋巴细胞 ,并呈现出较好的剂量效应关系。淋巴细胞凋亡是EMP引起淋巴细胞死亡和机体免疫功能降低的主要原因 ,Bax和Bcl 2在上述凋亡调控中起着重要作用。
Objective To study the injury effect of electromagnetic pulse (EMP) on canine lymphocytes and to explore its mechanism. Methods T, TH and TS lymphocytes were detected by acid phosphatase cytochemical method. The apoptosis of lymphocytes and the changes of Bcl 2 and Bax protein were analyzed by in situ end labeling and immunocytochemistry. Results The field intensity of (2 - 12) × 10 4 V / m decreased the number of T cells and their subpopulations in peripheral blood. The number of apoptotic lymphocytes increased significantly with the increase of irradiation dose. After 10 days of irradiation, The expression of Bax protein in lymphocytes was significantly increased, showing a good correlation with lymphocyte apoptosis, while the Bcl 2 protein was significantly decreased. Conclusion It was confirmed for the first time that EMP at a certain field strength can indeed damage lymphocytes and show a good dose-response relationship. Lymphocyte apoptosis is the main cause of EMP-induced lymphocyte death and decreased immune function. Bax and Bcl 2 play an important role in the above-mentioned apoptosis regulation.
