目的:探讨信号转导和转录激活因子-3(STAT3)基因rs2293152位点多态性与河南汉族人群肥胖易感性的关系。方法:采用整群抽样的方法收集河南省某农村地区汉族肥胖病例379人(病例组)和体重正常对照391人(对照组),应用限制性酶切片段长度多态性分析STAT3基因多态性;采用logistic回归模型分析基因多态性与肥胖的关联强度;采用MDR模型分析基因-环境之间的交互作用。结果:病例组和对照组STAT3基因rs2293152位点(CG+CC)和GG基因型频率分布差异有统计学意义(χ2=4.712,P=0.029)。调整年龄、性别等因素后,rs2293152位点(CG+CC)基因型携带者肥胖危险性升高(调整OR=1.391;95%CI:1.009~1.919)。基因-环境交互作用分析显示STAT3基因rs2293152位点和吸烟存在基因-环境交互作用(P<0.05)。结论:STAT3基因rs2293152位点多态性与肥胖有关联,且该基因多态性和吸烟对肥胖的发生具有交互作用。
Objective: To investigate the relationship between rs2293152 polymorphism of signal transducer and activator of transcription 3 (STAT3) gene and susceptibility to obesity in Henan Han population. Methods: A total of 379 obese cases (case group) and 391 normal weight controls (control group) were collected from a population of Han nationality in a rural area of Henan Province by cluster sampling method. The polymorphism of STAT3 gene was analyzed by restriction fragment length polymorphism Logistic regression model was used to analyze the association between gene polymorphism and obesity. The MDR model was used to analyze the gene-environment interaction. Results: There was significant difference in frequency distribution of STAT3 gene rs2293152 (CG + CC) and GG genotype between case group and control group (χ2 = 4.712, P = 0.029). After adjusting for age, sex and other factors, the risk of obesity was increased in carriers with rs2293152 (CG + CC) genotype (adjusted OR = 1.391; 95% CI: 1.009-1.919). Gene-environment interaction analysis showed that there was a gene-environment interaction between rs2293152 of STAT3 gene and smoking (P <0.05). Conclusion: rs2293152 polymorphism of STAT3 gene is associated with obesity, and the gene polymorphism and smoking have an interaction on the occurrence of obesity.
