胡黄连(Picrorhiza kurrooa)根的粉末以乙醇提取,提取液在50℃下真空蒸发,得到的残渣用苯提取。将不溶性部分溶于乙醇溶液中,部分以丙酮-乙醚沉淀,此步骤重复三次。将从丙酮-乙醚可溶性部分得到的组分溶于沸甲醇中,热可溶住部分经活性炭处理,过滤后冷藏,几天后即析出胡黄连素。将得到的胡黄连素上硅胶柱,用逐渐增加甲醇的乙酸乙酯溶液(1%~10%)
The powder of the root of Picrorhiza kurrooa was extracted with ethanol, the extract was evaporated in vacuum at 50° C., and the resulting residue was extracted with benzene. The insoluble fraction was dissolved in an ethanol solution and partially precipitated with acetone-ether. This step was repeated three times. The components obtained from the soluble portion of acetone-ether are dissolved in boiling methanol, and the hot-soluble portion is treated with activated carbon, filtered, and then refrigerated. After a few days, the berberine is precipitated. The obtained berberine was applied to a silica gel column with a gradually increasing methanolic ethyl acetate solution (1% to 10%).
