目的研究内皮型一氧化氮合酶(eNOS)基因多态性与2型糖尿病肾病(DN)的关系。方法选择207例(其中69例2型糖尿病肾病患者,75例2型糖尿病无肾病患者,63例正常健康对照者)为观察对象,从全血中提取DNA,进行聚合酶链式反应(PCR)并结合电泳分离技术检测eNOS基因4b/4a的多态性。结果 2型糖尿病肾病组a等位基因频率明显高于无肾病的2型糖尿病组和正常健康对照组(分别为χ2=8.2439,P<0.0167;χ2=7.6537,P<0.0167)。结论携带a等位基因的2型糖尿病患者比携带b等位基因更易罹患肾病,a等位基因可能是2型糖尿病肾病的一个独立危险因素。
Objective To study the relationship between polymorphism of endothelial nitric oxide synthase (eNOS) gene and type 2 diabetic nephropathy (DN). Methods Totally 207 patients (69 patients with type 2 diabetic nephropathy, 75 patients with type 2 diabetes without nephropathy and 63 normal controls) were selected as experimental subjects. DNA was extracted from whole blood and subjected to polymerase chain reaction (PCR) Combined with electrophoretic separation technology to detect eNOS gene 4b / 4a polymorphism. Results The frequency of a allele in type 2 diabetic nephropathy patients was significantly higher than that in type 2 diabetic patients without nephropathy and normal controls (χ2 = 8.2439, P <0.0167, respectively; χ2 = 7.6537, P <0.0167). Conclusion Type 2 diabetes patients with a allele are more susceptible to kidney disease than those who carry the b allele. The a allele may be an independent risk factor for type 2 diabetic nephropathy.
