不同空间排列静电纺丝对RSC96细胞表达NGF及Laminin的研究

目的研究不同空间排列静电纺丝对雪旺细胞株(RSC96)神经营养因子(NGF)及层粘连蛋白(Laminin,LN)的表达情况。方法根据培养条件不同分3组,即平行静电纺丝培养组(A)、随机静电纺丝培养组(B)及膜上培养组(C)培养3天后用Trizol法提取细胞总RNA,用Oligo(d T)逆转录酶得到c DNA。经特定引物PCR扩增,取相同量的逆转录产物进行q PCR反应,检测NGF及LN的表达差异。结果平行静电纺丝组,随机静电纺丝组,膜上培养组上NGF基因表达依次递增,各组间差异明显;LN基因的表达依次递减,各组间差异明显。结论不同空间排列的静电纺丝对RSC96细胞表达NGF和Laminin影响不同,平行的静电纺丝有促进细胞向成熟分化的可能。 Objective To study the expression of neurotrophic factor (NGF) and laminin (LN) in Schwann cell line (RSC96) with different spatial arrangements of electrospinning. Methods The total RNA was extracted by Trizol method after culturing for 3 days in different electrospun culture groups (A), random electrospun culture group (B) and membrane culture group (C) (d T) reverse transcriptase to obtain c DNA. After specific primers were amplified by PCR, the same amount of reverse transcription product was used for q-PCR to detect the difference of expression of NGF and LN. Results The expression of NGF gene in the parallel electrospinning group and the random electrospinning group increased gradually in each group, with significant difference among the groups. The expression of LN gene decreased in turn, with significant difference among the groups. Conclusion Electrospinning with different spatial arrangements has different effects on the expression of NGF and Laminin in RSC96 cells. Parallel electrospinning may promote the differentiation of cells into mature cells.

目的 探讨重症颅脑损伤患者复苏的治疗体会.方法 回顾性分析54例重症颅脑损伤患者的临床资料.结果 按格拉斯哥预后量表(GOS)评分标准,治疗后6个月评定结果:痊愈19例,占35.19%(19/54);轻残但生活能自理24例,占44.44%( 24/54);重残9例,占16.67%(9/54);死亡2例,占3.70%(2/54).结论 重症颅脑损伤患者进行早期复苏支持,是降低病死率、致残率的关键。
