外源性一氧化氮对骨肉瘤细胞株HOS生长能力的影响

目的 :研究外源性一氧化氮对骨肉瘤细胞株 (HOS)体外生长能力的影响。方法 :用不同浓度的SNP(Sodi umNitroprusside ,硝普钠 )作用于体外培养的HOS细胞株 ,经细胞作用产生外源性一氧化氮 ,通过Griess比色法、MTT比色法 ,观察HOS细胞株一氧化氮的产生量及对细胞株生长情况的影响。结果 :随着SNP浓度的增加 ,HOS细胞一氧化氮的产生量逐渐增加而细胞存活数量逐渐减少 ,以SNP 2 0 0 μmol/l浓度为一个分界点 ,2 0 0 μmol/L以下时变化不明显 ,2 0 0 μmol/L以上时变化明显 ,经统计学分析 ,2 0 0 μmol/L以上各组间差异明显 (P <0 .0 1)。结论 :一氧化氮对体外培养的骨肉瘤细胞株生长能力有一定的影响。 Objective: To investigate the effects of exogenous nitric oxide on the growth of osteosarcoma cell line (HOS) in vitro. METHODS: HOS cell lines cultured in vitro were treated with different concentrations of SNP (Sodium nitroprusside), and exogenous nitric oxide was produced by the cells. Griess colorimetric method and MTT colorimetric method were used to observe the changes of HOS cell line Nitric oxide production and its effect on cell growth. Results: With the increase of SNP concentration, the production of nitric oxide in HOS cells gradually increased and the cell survival number decreased gradually. When the concentration of SNP was 200 μmol / l, the change of NO was less than 200 μmol / L Obviously, the changes were obvious when the concentration was above 200 μmol / L. The difference between the groups above 200 μmol / L was statistically significant (P <0.01). Conclusion: Nitric oxide can affect the growth of osteosarcoma cells in vitro.
