为了探讨免疫抑制药物对同种异体带瓣主动脉移植的免疫排斥反应和远期疗效的影响,随机选取健康成年SD大鼠24只,分为实验组和对照组,分别接受同种异体带瓣主动脉腹主动脉的异位移植。实验组术后给予口服雷公藤多式片1.5mg/kg共2周。手术后2,4,8,12周测定外周血细胞ICAM-1和TCR-αβ的表达,并观察移植物的大体标本改变。实验结果显示:对照组术后早期(2~4周)ICAM司和TCR-αβ的表达明显增高,实验组术后2周TCR-αβ的表达和术后4周ICAM-1的表达少于对照组(P<0.01,P<0.05),二级移植物大体形态改变无明显差异。结论:雷公藤抑制了同种异体带瓣主动脉移植后机体的免疫反应。
To investigate the effect of immunosuppressive drugs on immune rejection and long-term efficacy of allogeneic aortic allograft transplantation, 24 healthy adult Sprague-Dawley rats were randomly selected and divided into experimental and control groups and received allogeneic flaps. Ectopic transplantation of aortic abdominal aorta. The experimental group received oral administration of 1.5 mg/kg Tripterygium wilfordii for 2 weeks. The expressions of ICAM-1 and TCR-αβ in peripheral blood cells were measured at 2, 4, 8 and 12 weeks after operation, and the gross specimen changes of the grafts were observed. The experimental results showed that the expression of ICAM and TCR-αβ was significantly increased in the early postoperative period (2-4 weeks) in the control group, and the expression of TCR-αβ at 2 weeks and the expression of ICAM-1 at 4 weeks after surgery were lower in the experimental group. In the group (P<0.01, P<0.05), there was no significant difference in the general morphology of the secondary grafts. CONCLUSION: Tripterygium wilfordii inhibited the immune response of allograft aortic allograft.
