目的研究抑郁大鼠脑内N-甲基-D-天冬氨酸(NMDA)受体亚单位NR1mRNA、NR2BmRNA的表达及中药颐脑解郁方的干预作用。方法二级雄性大鼠分为正常组、抑郁组、治疗组、对照组,后3组采用慢性应激方法结合孤养法建立抑郁模型,治疗结束后,取脑组织,采用逆转录PCR(RT-PCR)实验方法,研究NR1mRNA、NR2BmRNA在抑郁模型大鼠脑内左前额皮质、海马的表达及中药颐脑解郁方的干预作用。结果抑郁组大鼠海马、前额皮质的NR1、NR2B表达明显高于正常组,中药颐脑解郁方干预后可明显降低NR1mRNA、NR2BmRNA在脑内的表达。结论抑郁模型大鼠脑内NR1mRNA、NR2BmRNA表达升高,中药颐脑解郁方可使之明显下调,中药颐脑解郁方的抗抑郁作用可能与下调NMDA受体(NMDAR)的表达有关。
Objective To investigate the expression of N-methyl-D-aspartate (NMDA) receptor subunits NR1mRNA and NR2BmRNA in the brain of depressive rats and the intervention effect of Yinao Jieyu Decoction. Methods The second-grade male rats were divided into normal group, depression group, treatment group and control group. The latter three groups were induced by chronic stress and solitary method to establish depression model. After treatment, brain tissues were taken out and reverse transcription PCR -PCR) to study the expression of NR1 mRNA and NR2B mRNA in the left prefrontal cortex and hippocampus of rats with depression model and the interventional effect of Yinao Jieyu recipe. Results The expressions of NR1 and NR2B in hippocampus and prefrontal cortex of rats in depression group were significantly higher than those in normal rats. The expressions of NR1 mRNA and NR2B mRNA in brain were significantly decreased after Chinese medicine Yinao Jieyu intervention. Conclusion The expression of NR1mRNA and NR2BmRNA in the brain of depression model rats is increased. The Chinese herbal medicine Yinao Jieyu can significantly reduce the antidepressant effect, while the anti-depression effect of Yinaojieyin recipe may be related to the down-regulation of NMDA receptor (NMDAR) expression.
