目的:观察丹酚酸B和丹皮酚不同配比组合对大鼠离体胸主动脉血管的舒张作用和急性心肌缺血再灌注损伤的保护作用。方法:采用基线等比增减设计法进行药物配比设计,观察不同配比组合对去甲肾上腺素、氯化钾诱导的大鼠收缩血管环的舒张效应;结扎心脏左冠状动脉前降支(LAD)制备急性心肌缺血再灌注损伤模型,观察不同药物组合对心电图(ECG)、心肌梗死面积(MIS)、血清丙二醛(MDA)、一氧化氮(NO)、超氧化物歧化酶(SOD)和肌酸激酶同工酶(CK-MB)的影响。结果:丹酚酸B和丹皮酚各配比组合对离体血管舒张效果明显高于模型组,可减少再灌注损伤造成的心肌梗死面积,降低缺血程度(∑-ST),提高血清中SOD活力和NO含量,降低MDA含量和CK-MB水平;不同组合以丹酚酸B和丹皮酚之比为3∶1药物组合效果最佳。结论:丹酚酸B和丹皮酚配比组合对大鼠血管具有一定的舒张作用以及对大鼠急性心肌缺血再灌注损伤具有较好的保护作用,丹酚酸B和丹皮酚之间可能存在一定的协同或相加作用。
OBJECTIVE: To observe the protective effects of different combinations of salvianolic acid B and paeonol on the relaxation of isolated thoracic aorta and acute myocardial ischemia-reperfusion injury in rats. Methods: Base on the ratio of increase and decrease of the design of drug ratio design, observe the different combinations of norepinephrine, potassium chloride-induced vasoconstriction in rat vasoconstriction effect; ligation of left anterior descending coronary artery ( (LAD) for acute myocardial ischemia / reperfusion (I / R) injury model. The effects of different drug combinations on ECG, MIS, serum MDA, NO and SOD SOD) and creatine kinase isoenzyme (CK-MB). Results: The combination of salvianolic acid B and paeonol could significantly reduce the vasodilatation in vitro, which could reduce the area of myocardial infarction caused by reperfusion injury and reduce the degree of ischemia (Σ-ST) SOD activity and NO content, decreased MDA content and CK-MB level; the best combination of 3: 1 ratio of salvianolic acid B and paeonol was obtained in different combinations. CONCLUSION: The combination of salvianolic acid B and paeonol has certain vasodilatory effects on rat blood vessels and has a good protective effect on acute myocardial ischemia-reperfusion injury in rats. Between salvianolic acid B and paeonol There may be some synergy or additive effect.
