目的研究槲皮素、放射线单独及联用对体外培养宫颈癌Hela细胞增殖、凋亡、迁移和侵袭的影响并探讨分子机制。方法槲皮素、放射线单独及联合作用Hela细胞后,MTT法检测细胞增殖,流式细胞法检测细胞凋亡,划痕法及Transwell法检测细胞迁移及侵袭,免疫印迹法检测MMP-9蛋白。结果槲皮素、放射线单独或合用均可对Hela细胞产生明显的抑制增殖和诱导凋亡作用(P<0.05),联用效果更好;放射线可刺激Hela细胞迁移及侵袭并上调MMP-9蛋白表达,而槲皮素则可抑制这种作用并下调MMP-9蛋白表达。结论槲皮素可通过诱导凋亡方式增强宫颈癌细胞对放射线敏感性,并可通过下调MMP-9蛋白表达抑制放疗期间癌细胞的侵袭和转移。
Objective To investigate the effects of quercetin and radiotherapy alone and in combination on the proliferation, apoptosis, migration and invasion of cervical cancer Hela cells in vitro and to explore the molecular mechanisms. METHODS: Hela cells were treated with quercetin and radiotherapy alone. Cell proliferation was detected by MTT assay. Apoptosis was detected by flow cytometry. Migration and invasion were detected by scratch assay and Transwell assay. MMP-9 protein was detected by immunoblotting. Results Both quercetin and radiation alone or in combination could significantly inhibit the proliferation and induce apoptosis of Hela cells (P <0.05), and the combined effect was better. Radiation could stimulate the migration and invasion of Hela cells and up-regulate the expression of MMP-9 Quercetin inhibits this effect and down-regulates MMP-9 protein expression. Conclusions Quercetin can enhance the radiosensitivity of cervical cancer cells by inducing apoptosis and inhibit the invasion and metastasis of cancer cells during radiotherapy by down-regulating the expression of MMP-9.
